PD_AS_RNASeq

Analysis of RNA sequencing of alpha synuclein treated undifferentiated THP-1 monocytic cells.

Conditions

  • 5x105 cells/well in 6 well dish. 2ml of media -> 2.5x105 cells/ml
  • 24-hr treatment
  • Treated with WT monomeric (ASM) or preformed fibrillar (PFF) alpha synuclein
    • ASM 10μg/ml
    • PFF 1 and 10μg/ml

RNA Processing

  • RNA collected in 1ml TRIzol
  • TURBO DNase treated
  • Cold ethanol precipitated and suspended in 35μl nuclease-free H2O

Sequencing

  • polyA selection cDNA library prep
  • 2x150bp Illumina Hi-Seq performed by GENEWIZ
