Repository collecting parts lists and wiring diagrams for custom nematode thermotaxis rigs and a thermal stimulator for calcium imaging of imobilized worms.
This respository includes two thermotaxis rig versions:
- a large format arena (22x22 cm assay area) capabile of generating temperatures from 12 - 46 degrees Celsius. This version was first described in [Bryant et al 2018].(https://www.sciencedirect.com/science/article/pii/S0960982218306997) It is an updated implementation of the arena described in the [Wormbook chapter on Thermotaxis Naviation Behavior (see section 5)] (http://www.wormbook.org/chapters/www_thermonav/thermonav.html)
- a small format arena (~7.6x5 cm assay arena) that can generate temperatures in the C. elegans physiological temperature range, and may be capable of warmer temperatures. This version is an updated implementation of the arena described by Ramot et al 2018 and in the [Wormbook chapter on Thermotaxis Naviation Behavior (see section 2)] (http://www.wormbook.org/chapters/www_thermonav/thermonav.html)
The thermal stimulator for calcium imaging is designed for use on an upright Zeiss microscope.
