Missing Mutants and Molecular Biology about mwc_mutants HOT 2 CLOSED

Here is a list of the bad mutants as we discussed today

1. Y20I + Q294V (probably all operators)
2. Y20I + Q294K (probably all operators)
3. Q21A (probably all operators)

We also discussed putting all of the DNA binding domain mutants into the various RBSs (@mrazomej) that we used in the induction paper. This means that we will need to make the plasmids using the appropriate RBSs. This should be possible using only one primer pair.

• Integrate Q294R into O1 and O2 final strains (@nbellive)
• Sequence Q294R in O1 and O2 final strains (@nbellive) (temporary: In box 5, next to new O2 operator mutants)
• Need to reintegrate Q294K, F164T, into both operators @mrazomej , @gchure
• Need to transduce Q294V into O1 (@nbellive) (temporary: In box 5, next to new O2 operator mutants)
• Sequence Q294V in O1 and O2 final strains (@nbellive)
