Ah no, the Wine prefix was somehow set to Windows 7. After switching it to Win10, the game launches.

The problem with the Riot launcher is that it has a tendency to minimize itself in the system tray. And Ubuntu's interface (Gnome in general) handles system tray icons very poorly, especially when it comes to Windows applications.
I'm on KDE right now, I don't have issues un-minimizing the Riot Launcher. I don't know if you could give this a try?
There are also some Gnome Shell extension that make system tray icon better on Gnome, but in any case, I've never seen tray icons work well on Gnome no matter what extension is installed.
