You must call autoload function after the initialization of vim-plug.

set nocompatible

call plug#begin('~/.vim/plugged')
Plug 'liuchengxu/vim-which-key', { 'on': ['WhichKey', 'WhichKey!'] }
autocmd User vim-which-key call which_key#register('<Space>', 'g:which_key_map')
call plug#end()

let g:mapleader = " "
let g:maplocalleader = ","
nnoremap <silent> <leader>      :<c-u>WhichKey '<Space>'<CR>
nnoremap <silent> <localleader> :<c-u>WhichKey  ','<CR>

let g:which_key_map = {}

Otherwise if you don't know to how to utilize the on-demand feature of vim-plug, don't load vim-which-key on demand.

set nocompatible

call plug#begin('~/.vim/plugged')
Plug 'liuchengxu/vim-which-key'
call plug#end()

let g:mapleader = " "
let g:maplocalleader = ","
nnoremap <silent> <leader>      :<c-u>WhichKey '<Space>'<CR>
nnoremap <silent> <localleader> :<c-u>WhichKey  ','<CR>

let g:which_key_map = {}

call which_key#register('<Space>', 'g:which_key_map')
